The Museum of Old and New Art's event will take place from January 13 to 18 in Hobart.
Led by the Wayne Coyne, The Flaming Lips will play on the outdoor lawn stage at the Moorilla-based MONA.
The full Mofo line-up is set to be announced on October 16.
The Flaming Lips, from the US, was established in 1983. The band has quickly made a reputation for itself through its eclectic psychedelic rock tunes.
The band is also renowned for its live shows, where frontman Coyne performs inside a "space bubble" (a zorb ball) and the stage is littered with back-up dancers in furry animal suits.
