The two-vehicle crash on Boyer Road occurred about 12.30pm.
Police say that the deceased man was the sole occupant of a silver Nissan Patrol 4WD station wagon, which was travelling in a westerly direction.
It collided head-on with a black Ford Falcon sedan, which was being driven in the opposite direction by a 35-year-old woman from Conningham.
Police say the crash occurred on a straight section of Boyer Road and although the road was wet, it was not believed to be raining at the time.
The woman was taken to the Royal Hobart Hospital in a serious condition.
Investigating officer Sergeant Rod Carrick would like to hear from any member of the public who may have witnessed the crash or saw either vehicle beforehand.
