Ms O'Connor will lead Franklin MHA Nick McKim and Bass MHA Andrea Dawkins in the party room.
She promised the party would be a ``force to be reckoned with'' in its term of government.
``The task ahead is to continue to stand up for this beautiful island and its people, to articulate a vision for a prosperous, sustainable future, to hold government to account and build the Green vote across Tasmania,'' she said.
``We want to continue to deliver Green outcomes and to be recognised as much for what we propose as what we oppose.''
The decision came after the unexpected resignation of former leader Kim Booth last month.
Ms Dawkins was elected to Bass on countback and will deliver her inaugural speech when Parliament resumes Tuesday.
