The room is part of two new spaces at the St John Street venue, and has taken around six months to complete.
''Our biggest problem has always been not having enough space, as the business has grown much quicker than we expected,'' co-owner Ryan Campling said.
''A lot of people have also approached us for a private room for a function or a party, and it will be fantastic to offer this to people.''
The new spaces will take its official capacity to 200.
The Barrel Room will be opened for the first time from 3pm Saturday as part of the weekend-long Saint John Craft Beer's Paddock to Pot micro-festival.
It will open on Friday nights in the short-term after that.
