It will host DJ act Hot Dub Time Machine to celebrate the rebirth, with those in charge hoping to attract a touring band or DJ to the venue every six weeks.
The Hotel Tasmania has recently undergone renovations, removing the bistro to take it back to its former identity as a music venue so it can have a primary focus of entertainment. It has also dropped The Saloon name.
Manager Matt Harris yesterday said he believed now was the time for the business to again start hosting live music having not had a gig for the past few years.
"We worked out that there aren't many venues around here that do live music," Mr Harris said.
"I'm quite excited to get the room back to its former glory and we're really keen to get some live music back here.
"The fact that you can go somewhere else and listen to well known bands [and DJs] live, and get the whole band experience back."
Hot Dub Time Machine is DJ Tom Loud who starts with music from the 1950s and "mashes" up songs chronologically up until the modern day.
Doors open at 10pm, with tickets costing $20 from tixtas.com.au or $25 on the door.
The Hotel Tasmania's re-emergence as a music venue follows Club 54 in Cameron Street opening last year.
Lonnies Nightclub in Brisbane Street, which previously hosted live music, is also set to reopen soon.
