UPDATE 10.50am: It is believed the driver of the mini-van fell asleep at the wheel, police say.
The van was travelling from Claremont to Launceston for a religious gathering.
Senior Constable Sam Lloyd at Evandale Police said the driver was believed to have fallen asleep while driving, and woken up as the vehicle was leaving the road.
His reaction caused the car the roll, and it came to rest down an embankment.
The crash happened about 10 kilometres south of Woolmers Lane about 9.20am.
One woman was thrown from the car, and the driver had to be rescued by emergency services.
Both have been taken to the Launceston General Hospital in an unknown condition.
It is believed the remaining three passengers in the Kia Carnivale are believed to be relatively unharmed.
UPDATE 10.30am: Emergency services had to cut the roof off a mini-van to free its passengers after the vehicle crashed on the Midland Highway.
It is not known how many people were in the vehicle, but the van was believed to be travelling as part of a group.
It was driving towards Perth.
EARLIER: Police have advised that the Midland Highway at Perth is closed to traffic after a crash.
It is a single-car crash where the car has rolled, trapping its occupants.
Injuries are not known at this stage.
Motorists are asked to divert from the highway at Woolmers Lane, through Cressy, and onto Powranna Road.
