Dunk scored a record 229 not out for Tasmania at North Sydney Oval at the weekend.
The powerful opener's selection has seen him leapfrog fellow Tasmanian Tim Paine as well as NSW keeper Peter Nevill, WA's Sam Whiteman and former Tasmania and Australian wicketkeeper Mat Wade for the top role.
Regular Australian keerp Brad Haddin will still be in the United Arab Emirates playing Test cricket against Pakistan when the three-match series kicks off against South Africa on November 5.
Dunk joins fellow Tiger James Faulkner, whose selection is subject to his requirements on the tour of the UAE.
