Police say the men committed the armed robbery just after 10pm.
The men forced two staff to hand over an undisclosed amount of cash before taking off.
The culprits, armed with a wooden stick and what appeared to be a pistol, entered the hotel via the rear door, police said.
The incident was captured on CCTV, but both men had their faces covered.
Police would not comment on suggestions the staff had been locked in a safe during the robbery, which occurred as the night's takings were being counted.
Yesterday the usually busy pub was shut with the publican declining to comment.
Detectives from Launceston CIB are investigating and have door- knocked surrounding businesses.
Police are seeking assistance from members of the public who may have seen anyone acting suspiciously before the robbery.
Anyone with information can call Crime Stoppers anonymously on 1800 333 000.
