The new spillage was at a manhole on the boundary of 6 Esplanade, Campbell Town.
A TasWater spokeswoman said the company became aware of the spill at 10am and the overflow was stopped at 11.45am.
"Warning signs were erected and samples were taken upstream and downstream of the point of overflow," the spokeswoman said.
"The area affected has been cleaned up and limed.
"Results of samples will be provided when they are available."
The spokeswomen said a downstream user, about one kilometre from the overflow was notified of the spill by phone yesterday afternoon.
"Our priority is at all times to minimise the impact on the surrounding environment," the spokeswoman said.
"The relevant regulatory authorities were notified as part of standard procedures followed in such instances."
