The 32-year-old Chinese National was the passenger in a car being driven by her 30-year-old husband.
The pair were touring the state.
About 11.25am on the Bass Highway west of Port Latta, a white Hyundai delivery van collided with the couple's silver Volkswagen hatch.
Both vehicles came to rest down an embankment.
The driver of the Volkswagen was trapped in the car and had to be freed by the Circular Head SES road rescue team.
The husband and wife were taken to the North West Regional Hospital at Burnie.
The woman died from her injuries about 4.30pm, while her husband is in a serious condition after surgery.
The 31-year-old driver of the delivery van, who was from Burnie, was admitted and released from hospital with minor injuries.
Anyone who witnessed the crash or saw either vehicle before the crash is urged to contact police.
Police reminded motorists to obey the road rules, remain vigilant when driving and take rest stops on long journeys.
