Police said the woman was travelling south on a black 250cc Suzuki when she lost control and crashed in to an Armco railing at 10.38am.
According to police, high winds are believed to have been a factor in the crash.
Police say speed and alcohol were not involved.
The annual toy run, organised by the South Motorcycle Riders Association, departed from Perth Roadhouse mid-morning.
The riders were travelling to the Parliament House lawns in Hobart, where they were to hand over gifts, food, toys and donations to the Salvation Army.
